چکیده

Several analogues of the posterior pituitary hormones, oxytocin I and the vasopressin# II and III, have been synthesized in this laboratory (l-5) in an attempt to detect possible correlations between chemical structures and biological properties in this group of compounds. Among these analogues is oxypressin IV (l), a cyclic octapeptide amide which contains a cyclic pentapeptide amide portion identical with the one existing in the vasopressins, linked to the tripeptide amide side chain that is present in oxytocin. It may be recalled that oxypressin possesses very low pressor activity, although it contains the ring structure that is present in the vasopressins. This may be attributable (2) to the absence of a strongly basic amino acid in the side chain, such as is present in the vasopressins. It was therefore of interest to synthesize and determine the biological properties of a compound which, like oxypressin, would also be a combination of the two moieties existing in osytocin and vasopressin but, unlike oxypressin and histidinevasopressin, would have a strongly basic amino acid in the side chain.

Lysine-vasotocin, a synthetic analogue of the posterior pituitary hormones containing the ring of oxytocin and the side chain of lysine-vasopressin.

The synthesis of oxytocin (I), arginine-vasopressin (2)) and lysine-vasopressin (3-5) has opened the way for the synthesis of analogues of the natural hormones, thus affording an opportunity to study the relationship between structure and biological activity in these hormones. One of the most interesting analogues thus far prepared is arginine-vasotocin, an octapeptide amide containing the ring...

Identification of neurohypophysial hormones with their antisera.

Identifying posterior pituitary hormones in body fluids or neurohypophysial extracts was heretofore partially achieved by using pharmacologic potency ratios or semispecific inactivation by thioglycolate or enzymes. Production of antisera against oxytocin and lysine-vasopressin has prompted us to test their specificity against lysine-vasopressin, arginine-vasopressin, arginine-vasotocin, and oxy...

Conformational studies on arginine vasopressin and arginine vasotocin by proton magnetic resonance spectroscopy.

The assignments and partial conformational analysis of the 220-MHz (1)H nuclear magnetic resonance spectrum of the neurohypophyseal hormones arginine vasopressin and arginine vasotocin in uniformaly deuterated dimethylsulfoxide are reported. The chemical shifts, vicinal coupling constants, temperature coefficients of chemical shifts, and proton-deuterium exchange rates for protons of arginine v...

Influence of the peptide-chain length on disulphide-bond formation in neurohypophysial hormones and analogues.

(8-Arginine)vasopressin, (8-arginine)vasotocin, oxytocin and oxypressin, the 'ring' derivatives pressinamide and tocinamide, and the extended-chain analogues Pro-Arg-Val-(8-arginine)vasopressin and (8-arginine)vasopressinoyl-Ala-Met-Ala-NH(2), were synthesized by the solid-phase method and purified by sequential gel filtration on Sephadex G-15 in 50% acetic acid and 0.2M-acetic acid. Controlled...

Posterior Pituitary

Vasopressin and oxytocin are the only hormones known to be secreted by the neurohypophysis in humans. Human vasopressin is called arginine vasopressin (AVP) to distinguish it from lysine vasopressin in pigs. It is also called antidiuretic hormone (ADH), which reflects its major physiologic action. These hormones are nonapeptides (nine amino acids) with similar primary structures but very differ...
